Enzyme immunoassay for the determination of antibodies against Helicobacter pylori in human serum.
Helori-test IgG Chromo can be used for the qualitative determination of lgG specific antibodies directed against Helicobacter pylori in the human serum, which have a clinical significance in the gastric pathologies. Helori-test lgG Chromo represente a support to be used togheter with the clinical data for the diagnosis of this disease.
PRINCIPLE OF THE PROCEDURE
Helori-test lgG Chromo is a sandwich type enzyme-immunoassay. The purified antigen is attached to microtitration wells. Antigen-specific antibodies in the patient's serum bind to the antigens. Non-specific antibodies are removed by washing.
Alkaline phospahatase-labelled sheep-antihuman lgG are added and bind to human antibodies in the well. The excess conjugate is washed away; then a chromogenic substrate is added. After an appropriate incubation period, the Optical Density of the substrate is measured by means of a EIA reader. The higher the amount of specific antibodies present in the sample, the greater the O.D. values.
TEST PROCEDURE
- Pipette sample diluent, positive and negative control and diluted samples
- incubate for 60 minutes at 37 °C
- Wash 3 times with the washing solution
- Pipette conjugate in each well
- Incubate for 60 minutes at 37 °C
- Repeat washing steps as above
- Pipette substrate in each well
- Incubate for 30 minutes at 37 °C
- Stop the reaction by adding stop solution
- Read the results at 405 nm
