Helori AST is an enzyme immunoassay based on labelled monoclonal antibodies. This feature allows to reach a very high specificity still having a very high sensitivity. In this way, the method is a good as the urea breath test in the monitoring of eradaction therapy with antibiotics and can be run just using the normal laboratory equipment.

Microtiter strip wells are pre-coated with a mixture of monoclonal antibodies against H.pylori antigens. Diluted stool samples and controls are pipetted into the wells with mixture of monoclonal antobodies cojugated to horseradish peroxidase and incubated for further 90 minutes, at room temperature. The fecal H.pylori antigens will be sandwiched between the solid phase and enzyme-linked antibody.

After washing the wells, the immune complex formed by the bound conjugate is visualized with TMB sbstrate which gives a blue colour. The stop solution ends the reaction and turns the blue colour to yellow. The colour intensity is directly proportional to the amount of faecal antigen present in the sample.

START PROTOCOL

There are two steps :

  1. Extraction
  2. Reading the results on the plate following the below listed procedure:
  • Sample extraction
  • Controls and extracted sample addition
  • Incubate for 30 min. at RT (room temperature)
  • Addition of conjugated anti-Helicobacter-antibody
  • Incubate for 30 min. at RT
  • Washing step
  • Addition of TMB substrate
  • Incubate for 10 min at RT
  • Stop the reaction
  • Visual reading and/or at 450 nm
